Overview of Breeding Soundness Examination (BSE) of the Male Animal
Definition: A systematic evaluation of a male's reproductive potential and ability to get females pregnant.
• Limitation: It is not a direct evaluation of fertility, which can only be confirmed by the production of offspring.
• Initial Steps: Requires proper animal identification and a detailed history, especially for sub- or infertile males.
Primary Components of the BSE
• General Physical Evaluation:
◦ Physical examination and inspection of genital organs.
◦ Scrotal Circumference (Ruminants): Measured to estimate daily sperm output and "serving capacity" (the number of females a bull or ram can impregnate).
• Behavioral Assessment:
◦ Evaluation of breeding ability and libido.
◦ Note: This is difficult to assess during routine collections via electroejaculation.
• Breeder Classification:
◦ Satisfactory: Meets all criteria.
◦ Questionable: Potential issues found.
◦ Unsatisfactory: Assigned if the animal has inherited physical defects, such as cryptorchidism.
Semen Quality Evaluation:
• Gross Appearance: Visual inspection of the sample.
• Sperm Quantity: Calculation of total sperm in the ejaculate using volume and concentration.
• Sperm Motility:
◦ Gross motility (specific to ruminants).
◦ Individual motility (total and progressive) of raw or diluted samples.
• Morphology & Cytology:
◦ Percentage of morphologically normal sperm.
◦ Evaluation for hemospermia (red blood cells) or pyospermia (white blood cells).
Ancillary (Advanced) Sperm Tests
• Membrane Integrity:
◦ Evaluated using Eosin stain (non-viable sperm turn pink/red) or fluorescent probes like SYBR-14 and propidium iodide.
• DNA Integrity:
◦ Sperm Chromatin Structure Assay (SCSA): Uses acridine orange to detect if sperm DNA can withstand acid denaturation.
• Functional Assessments:
◦ Tests for capacitation, acrosomal exocytosis, and the ability to bind to or penetrate the oocyte.
• Constraints: These tests require high expertise and specialized facilities and are rarely used in routine clinical BSEs.
